abstract | The invention discloses a method for revealing an action mechanism of miRNA on liver cancer by using Small RNA sequencing technology, which comprises the following steps: step S1, extracting total RNA of liver cancer tissues and paracancer normal tissues thereof, and then carrying out sample RNA quantity, purity and integrity detection; step S2, performing reverse transcription and amplification on qualified RNA to construct a cDNA library, and detecting a Small RNA sequence of a sample by an Illumina Small RNA high-throughput sequencing technology; step S3, performing quality control processing, sequence comparison, sequence classification and new miRNA prediction operation on the sequencing result to obtain the information of known miRNA and new miRNA; step S4, normalizing the miRNA expression quantity, and predicting the miRNA target gene with differential expression; step S5, performing subsequent gene ontology GO biological function and KEGG channel analysis on the differential expression target genes. The miRNA and the related biological information thereof obtained by the invention have important significance for liver cancer research, and the related miRNA is a potential biomarker for liver cancer diagnosis and prognosis. |
