abstract | The invention discloses the nucleic acid sequence of a pair of human monoclonal antibodies that specifically bind to Mycobacterium tuberculosis complex (MTBC) lipoarabinomannan (LAM), and the preparation method and application of the recombinant antibody. The monoclonal antibody disclosed in the present invention is obtained by screening and identification of healthy human scFv phage antibody library inoculated with BCG, and its antibody sequence is obtained by sequencing, and the antibody sequence is further cloned into an expression vector, and then transfected into 293 cells and purified. The recombinant antibodies prepared by the invention can be combined with MTBC LAM with high sensitivity. Based on the monoclonal antibody pairing, an immunochromatographic reagent card for rapid detection of LAM antigen is further developed, which can specifically detect MTBC and some slow-growing mycobacteria lysates. , and does not cross-react with fast-growing mycobacteria. The immunochromatographic reagent card developed by the present invention can be directly used for qualitative (taking colloidal gold reagent card as an example) or quantitative detection (taking quantum dot fluorescent microsphere test paper card as an example) of urine samples of tuberculosis patients, and has a high performance. The detection sensitivity and specificity are extremely high, the operation is simple and time-consuming, and it has broad clinical application prospects in the rapid diagnosis of tuberculosis. |
