abstract The invention provides a method for preparing a blocking reagent of a repetitive sequence by a multiple substitution amplification technology, which comprises the following steps: amplifying the genome DNA by adopting a multiple substitution amplification technology; after purifying the amplification product, breaking the DNA fragment with the length of 200-300bp, and obtaining the product which is the blocking reagent of the repetitive sequence. The method for preparing the blocking reagent of the repetitive sequence can overcome the defects that the placenta DNA is not easy to obtain and the HAP column separation method is complicated, time-consuming and labor-consuming in the prior art.
