According to the invention, through a site-directed mutagenesis method, lysine at the 15 th site of the acetylacetone lyase is changed into glutamine, alanine at the 60 th site of the acetylacetone lyase is changed into aspartic acid, and after the acetylacetone lyase mutant is used for synthesizing acetylacetone, the yield of acetylacetone is improved to 112.9 mg/L.
