abstract | The invention provides a synthesis method of a pexidinib intermediate, which specifically comprises the following steps: taking 6-amino nicotinic acid ester compounds and 6-trifluoromethyl nicotinaldehyde as raw materials to synthesize compounds I, taking the compounds I as raw materials, and carrying out reduction reaction by using a reducing agent to generate [6- ({ [6- (trifluoromethyl) pyridin-3-yl ] methyl } amino) pyridin-3-yl ] methanol; the [6- ({ [6- (trifluoromethyl) pyridin-3-yl ] methyl } amino) pyridin-3-yl ] methanol is used as a raw material and is oxidized by an oxidant to generate 6- ({ [6- (trifluoromethyl) pyridin-3-yl ] methyl } amino) pyridine-3-formaldehyde. The synthesis method has the advantages of cheap and easily-obtained raw materials, simple and convenient operation, mild reaction conditions, high yield (the total yield is more than 58.62 percent) and the like, can realize large-scale production, and reduces the production cost of the pexidininib, thereby improving the market competitiveness of the product. |
