The invention belongs to the technical field of liver diseases, and in particular relates to a group of shRNA and cDNA targeting and regulating alpha-fetoprotein (AFP) gene and application thereof. The invention provides shRNA and cDNA for liver-targeted regulation of alpha-fetoprotein gene and application thereof. The shRNA-based recombinant vector provided by the present invention can silence the alpha-fetoprotein gene, and construct a model for reducing liver injury, endoplasmic reticulum stress and liver cell necrosis. After knocking down the Afp gene, the mouse liver injury and endoplasmic reticulum stress in the liver were aggravated. After overexpression of the Afp gene, liver injury and endoplasmic reticulum stress in the liver were alleviated in mice. It shows that reducing the level of AFP protein will aggravate the liver damage, which shows that the increase of intrahepatic AFP is beneficial to reduce liver damage, and can be applied to the preparation of drugs for reducing liver damage, protecting liver endoplasmic reticulum stress or treating liver cancer. |
