The invention adds the solid separation medium commonly used in the fixed bed column chromatography into the separation column of the countercurrent chromatography, and forms the state of the expansion and dispersion of the adsorption medium in the separation column based on the countercurrent chromatography fluid dynamics balance principle to construct the expanded bed countercurrent chromatography, which can be used as a novel chromatographic separation method for separating polar polysaccharide heparin drugs. The method has the advantages of high sample processing amount, remarkably improved utilization rate of the adsorption medium, and good application prospect in separation and purification of polar polysaccharide bioactive macromolecules. |
