abstract The invention relates to a method for judging the position of a double bond in unsaturated polymyxin, wherein the unsaturated polymyxin generates epoxidized polymyxin or/and dihydroxylated polymyxin (but not limited to the components, and may also comprise other polymyxin oxidation products, epimerization products, hydrolysis products, sulfation products, sulfonation products, methanesulfonic acid products and the like) under the illumination condition, and the position of the double bond in the unsaturated polymyxin can be judged by confirming the structure of the components through high performance liquid chromatography-high resolution mass spectrometry or/and high performance liquid chromatography-secondary mass spectrometry. The method can be used for judging the position of the unsaturated bond in the double-bonded polymyxin without component preparation, so that the reagent and labor cost are saved; the method for synthesizing and determining the structures of the epoxidized polymyxin and the bihydroxylated polymyxin is suitable for other polypeptide antibiotic components, simple and efficient and easy to popularize.
