abstract The invention discloses a method for dynamically correcting a DNA fluorescence spectrum by using a multicolor fluorescence internal standard, which comprises the following steps: determining an STR locus to be detected, and planning the arrangement of each fluorescent color channel in the STR locus; designing a multicolor fluorescence internal standard; during each DNA fragment detection and analysis, the earlier-embedded multicolor fluorescence internal standard is identified, and a corresponding spectrum correction matrix is dynamically constructed to correct the DNA fluorescence spectrum. By utilizing the invention, a spectrum correction reagent and an independently arranged spectrum correction link are not required to be specially equipped, DNA detection technicians do not need to pay attention to the correspondence of the spectrum correction matrix and the kit, and do not need to pay attention to the complicated problems of reworking and the like when the spectrum correction distortion needs to be carried out, so that the application complexity of the STR detection technology is obviously reduced, the detection cost can be undoubtedly reduced, and the STR detection method has good potential economic benefit and important application value.
