abstract The invention provides a novel Tafamidis derivative and a synthesis method for preparing the Tafamidis derivative from a catechol compound, ammonium acetate and an aldehyde compound by using sodium periodate as an oxidant. The method has the advantages of simple reaction conditions, low raw material synthesis cost, improved atom economy of reaction, reduced reaction steps, avoidance of the use of an acylation reagent of dichlorobenzoyl chloride and an esterification reagent of (trimethylsilyl) diazomethane, and solution of the problem of environmental hazard caused by acyl chloride, diazo reagents, strong acid and strong base additives and the like. Sodium periodate is used as an oxidant in the reaction, and iodate generated after the reaction is finished is insoluble in a reaction solution and can be well recycled, so that the pollution caused by a reaction system is greatly reduced, a more economic and environment-friendly path is provided for the synthesis of the novel Tafamidis derivative, and the application value is very high.
