abstract The invention provides a HEV-specific crRNA based on CRISPR-Cas12a technology, a detection kit and application thereof, and belongs to the technical field of hepatitis E virus (HEV) detection. The present invention provides HEV-specific crRNA including four g RNAs based on CRISPR-Cas12a technology. Experiments show that the four gRNAs in crRNA can specifically target HEV of four specific genotypes, and have high binding specificity. The invention provides a detection kit for HEV comprising HEV-specific crRNA, CRISPR-Cas12a protease and a single-stranded DNA reporter system. The present invention combines isothermal amplification technology with CRISPR-Cas12a technology. The detection limit of this method can reach 10copies/μl, and the specificity is good, which provides an accurate, fast and simple detection method for clinical diagnosis and laboratory research.
