titleOfInvention | Precisely engineered stealth messenger RNAs and other polynucleotides |
abstract | The present disclosure relates to methods for reducing immunogenicity in long polynucleotide sequences by precise sequence engineering of the immunogenic motifs in said polynucleotide sequences. The present disclosure also relates to precise sequence engineered polynucleotides with improved functionality such as exhibiting low innate immunogenicity, improved stability, or high protein expression. In these polynucleotides, immunogenic sequence motifs are removed, while the remainder of the sequence is retained. Compared to total nucleotide changes, this targeted engineering approach has unique advantages, including less disruption of native or optimized polynucleotide sequences, and thus maintaining high expression capacity, while being able to target innate immune receptors.
