The invention discloses a construction method of a sodium gluconate-producing Aspergillus niger genetically engineered bacteria overexpressing CAT and GOD, and belongs to the technical field of bioengineering. In the present invention, a recombinant plasmid comprising a cox promoter and a TtrpC terminator is first constructed, and then the CAT and GOD genes are connected through different linkers, and then introduced into the Aspergillus niger host strain to overexpress CAT and GOD at the same time, so as to realize the extracellular CAT and GOD of the engineered bacteria. A large amount of secretion, the final production of sodium gluconate at the shake flask level of the engineering bacteria can reach up to 20.5g/100mL fermentation broth, and the engineering strain in the 15L fermentation tank is fermented for about 18 hours, and the sodium gluconate yield can reach 34.7g/100mL fermentation broth. Compared with the wild-type strain FG‑2, the fermentation time of the engineered strain was shortened by 10%, laying a foundation for further improving the yield of industrially produced sodium gluconate. |
