abstract | The invention provides a method for separating and enriching circulating tumor DNA, which comprises the following steps: step 10), carrying out centrifugal treatment on the plasma sample to obtain a first supernatant; step 20) centrifuging the first supernatant to obtain a second supernatant; step 30) centrifuging the second supernatant to obtain a third supernatant; step 40) removing fibrin in the third supernatant to obtain a fourth supernatant; and 50) removing exosomes in the fourth supernatant to obtain a fifth supernatant. According to the separation and enrichment method of the circulating tumor DNA, cells and large fragments can be removed after the first centrifugal treatment, blood platelets can be removed after the second centrifugal treatment, large vesicles can be removed after the third centrifugal treatment, fibrin can be removed for the fourth time, exosomes can be removed for the fifth time, and the content of the circulating tumor DNA in the finally obtained fifth supernatant is higher than that of the circulating tumor DNA obtained by the existing method. |
