abstract | The invention discloses a cross-linked agarose affinity medium and a preparation method and application thereof. The preparation method includes the following steps: (1) dispersing agarose-containing dispersed phase and emulsifier-containing continuous phase in a microchannel reaction device (2) the agarose microspheres obtained in step (1) are sequentially processed through the following steps to obtain a cross-linked agarose affinity medium; (i) swelling , activation, polyhydroxy polymer modification; (ii) cross-linking; (iii) activation, coupling iminodiacetic acid ligands; (iv) chelating nickel ions. The modified microspheres are used for the purification and immobilization of enzymes, which solves the problems of easy inactivation and inability to recycle and reuse the enzymes in the complex separation and purification process. The influence of the miscellaneous enzymes in the enzyme solution on the product was investigated, and the method for preparing cytidine has the advantages of mild reaction conditions, green environmental protection and the like. |
