abstract | A hybridoma cell line secreting diphenhydramine monoclonal antibody and application thereof belong to the field of cosmetic safety immunodetection. The hybridoma cell line SHS 5B11 that secretes the diphenhydramine monoclonal antibody of the present invention is classified and named as a monoclonal cell line, and is deposited on September 27, 2020, with the deposit number CGMCCNo. The monoclonal antibody secreted by this cell line has good specificity and detection sensitivity to diphenhydramine. In the present invention, the complete antigen of diphenhydramine is synthesized, mixed and emulsified with an equal amount of Freund's adjuvant, immunized BALB/c mice by subcutaneous injection on the back, screened by indirect competitive enzyme-linked immunization method and subcloned three times to obtain Hybridoma cell line SHS 5B11. The monoclonal antibody secreted by this cell line has good detection sensitivity for diphenhydramine (IC 50 value is 2.93ng/mL), and can be used for the residual detection of diphenhydramine in cosmetics. |
