abstract | The invention discloses a novel coronavirus nucleic acid purification reagent and a purification method, belonging to the technical field of nucleic acid purification. The nucleic acid purification reagent comprises nano-magnetic beads coated with selenolysine-modified chitosan, a binding solution, and a washing I solution , wash II solution and eluent. The purification method includes: adding the sample to be purified, the magnetic beads and the binding solution into a centrifuge tube, mixing evenly, magnetically separating, and discarding the supernatant; adding washing I solution to the centrifuge tube, washing, magnetically separating, and aspirating the cap and bottom of the tube. Add the washing II solution to the centrifuge tube, wash, magnetically separate, and aspirate the residual liquid from the tube cover and the bottom of the tube; add the eluent to the centrifuge tube, place it at 60‑70 ℃ for 10‑15min, every Gently shake for 1-2 minutes to mix, magnetically separate, carefully pipette the supernatant into a new centrifuge tube, and store in a -20°C refrigerator. The method of the invention has rapid purification process, accurate results and high precision, and can improve the yield, purity, repeatability and stability of nucleic acid purification. |
