abstract | A method for enhancing cross presentation of dendritic cells by 27-bit lysine mutant ubiquitin molecules relates to the technical field of genetic engineering. Separating dendritic cells from the bone marrow of the mice; pre-treating bone marrow-derived dendritic cells with 27 th lysine mutant ubiquitin molecules: siRNA interference experiment is firstly carried out, then in-vitro adoptive immunity is carried out, and finally enzyme-linked immunospot experiment and immunoblotting experiment are carried out. The experiment proves that the dendritic cells treated by the ubiquitin molecules with the arginine mutated from the lysine at the 27 th position can obviously enhance the cross presenting capability of the dendritic cells and promote the antigen specificity CTL induction capability mediated by the DC; the ubiquitin molecule of which the 27 th lysine is mutated into the arginine can increase the capacity of the lymphocyte in the dendritic cell transfusion receptor to generate perforin and granzyme B. |
