abstract | The invention belongs to the field of LDLR gene research, and specifically discloses LDLR gene mutants and applications thereof. Included are nucleic acids with the c.2517-2518insCA mutation compared to the wild-type LDLR gene; and polypeptides with the p.C839fs mutation compared with the wild-type LDLR. It also relates to a construct comprising the aforementioned nucleic acid and/or the aforementioned gene mutation, and a method for preparing a plasmid for preparing a drug for preventing and treating hypercholesterolemia, comprising the steps of using the wild-type LDLR gene as a template to amplify the mutant LDLR cDNA Fragment, digest the GATC sequence methylated at the N6 position of adenine in the DNA double-strand, purify the mutant fragment, double-enzyme digestion and enzymatic linking of the mutant fragment and the expression vector to obtain the expression plasmid, prepare the plasmid bacteria liquid, and extract from the plasmid bacteria liquid plasmid. The present invention discovers a new mutation site of the LDLR gene, and the detection of the mutation site can be used for auxiliary diagnosis of hypercholesterolemia patients, and can be further applied to molecular diagnosis of hypercholesterolemia patients. |
