abstract The invention discloses a method and a kit for simultaneously detecting activity of aldosterone and renin in a blood sample, which comprises a sample pretreatment step and a sample injection detection step, wherein the sample pretreatment step comprises the following steps: adding an incubation buffer solution into a blood sample, incubating for a set time, adding a pH value regulator, purifying to obtain a treatment solution, drying the treatment solution by using nitrogen, and re-dissolving by using a mobile phase to obtain a solution to be detected; the pH value regulator is formic acid, ammonia water or ammonium acetate; and detecting by adopting a liquid chromatography-tandem mass spectrometry method. The optimization direction of the method is that on one hand, aiming at the physical property of angiotensin I, the pretreatment process is optimized, 10-100 mM ammonium acetate is used as a pH value regulator, so that the protonation dissociation of the angiotensin I in a solution to be detected is reduced, the recovery rate of the angiotensin I in SPE treatment is improved, and the detection sensitivity of substances is greatly improved; on the other hand, after SPE extraction, nitrogen blowing operation is added, and sample loading is carried out after redissolution, so that the detection signal is improved.
