abstract The invention discloses a genetically engineered bacteria, a plasmid combination, the use of the plasmid combination to prepare the genetically engineered bacteria and the use of the genetically engineered bacteria. The plasmid combination includes a cas9 expression plasmid and an sgRNA expression plasmid containing a heterologous proline hydroxylase gene, which can be used to prepare the genetically engineered bacteria of the present invention. Using the genetically engineered bacteria of the present invention, high-purity PB 0 can be prepared without adding proline and without purification, and no PC 0 by-product is produced, and has wide industrial application.
