abstract |
The present invention relates to methods of characterizing target DNA polynucleotides using Rolling Circle Amplification (RCA) and synthetic single stranded guide rna (sgrna) to identify and cleave the WT version of the target DNA polynucleotide. Also provided are characterization steps based on the use of a transmembrane pore and a DNA translocase that controls the movement of a DNA polynucleotide through the transmembrane pore. Further contemplated is a kit comprising one or more oligonucleotides specific for at least a portion of a target DNA polynucleotide, a sgRNA specific for a WT version of the target DNA polynucleotide, and a sgRNA-guided nucleic acid binding protein. |
