abstract | The embodiment of the invention provides a T cell PD-1 detection method based on cell mitochondrial quality evaluation, which comprises the following steps: s10: obtaining T cells; s20: adding a fluorescently-labeled monoclonal antibody into the T cells to coil out helper T cells and/or cytotoxic T cells; simultaneously adding a PD-1 antibody which is labeled by fluorescence to obtain a first sample; s30: adding a mitochondrial probe into the first sample, and dividing an MT-group and an MT + group by using the mitochondrial probe as a cell metabolism index to obtain a second sample; s40: detecting the second sample by using a flow cytometer to obtain the expression quantity of PD-1 under different metabolic levels; wherein the MT-group is a low-intensity expression group after the mitochondrial probe is stained, and the MT + group is a high-intensity expression group after the mitochondrial probe is stained. The technical scheme provided by the invention refines PD-1 detection expression through mitochondrial quality evaluation, so that the detection result is more accurate. |
