Aiming at the problems of transcription inhibition of purine operon pur and difficult accumulation of purine compounds in a purine de novo synthesis pathway, the invention provides a mutant for improving the expression level of purine operon pur gene, which is constructed by knocking out a DNA combined transcription two-component regulatory factor arcA (Genbank accession number is 948874) in escherichia coli. The invention realizes the regulation of the regulation factor arcA in purine operon pur gene expression and the application in purine biosynthesis for the first time.
