abstract The invention discloses a high-throughput screening method of a CRISPR/Cas9 library of brain-transition-related candidate genes, which comprises the steps of constructing a sgRNA library of the brain-transition-related genes, packaging the sgRNA library with lentiviruses, infecting host cells stably expressing CAS9 protein, screening to obtain cells stably expressing the sgRNA library, then inoculating the cells infected with the lentiviruses into an intracranial and fat pad of an immunodeficient mouse, finally, taking out tumor tissues with intracranial neoplasia and subcutaneous neoplasia, extracting genome DNA, performing second-generation sequencing on a targeting region of the sgRNA to build a library, and finding out the brain-transition-related candidate genes through the abundance change of the sgRNAs before and after screening. The method establishes a high-throughput screening technology of the brain transfer related gene, can accurately and efficiently excavate and control the key gene of brain transfer, and provides a new target and a new way for preventing and treating the brain transfer.
