titleOfInvention Natto kinase engineering bacterium and method for improving activity of natto kinase
abstract The invention provides a nattokinase engineering bacterium and a method for improving the activity of nattokinase, which comprises the following steps: step one, respectively extracting genome DNAs of bacillus subtilis and bacillus natto; secondly, designing a first primer pair and a second primer pair respectively for PCR amplification by taking a transcription regulation factor sinR and a nattokinase coding gene aprN as target genes and taking DNA of bacillus subtilis and bacillus natto as templates to obtain a sinR PCR product and an aprN PCR product; thirdly, constructing a recombinant plasmid of the sinR and the aprN in series connection by using the sinR PCR product and the aprN PCR product; step four, transforming the recombinant plasmid into competent cells to obtain a positive clone colony; and step five, sequentially carrying out seed culture medium culture and fermentation culture on the positive clone colonies. The invention can express the quorum sensing regulation factor and the nattokinase coding gene in series, thereby improving the nattokinase content and activity.
