abstract The invention discloses a method for enhancing the expression of a positive regulatory protein gene to improve the fermentation level of Jinggangmycin. By enhancing the expression of positive regulatory protein genes SHJG_1109, SHJG_1617, SHJG_5241 and SHJG_6272 in Streptomyces hygroscopicus using the strong promoter kasOp*, a high-yielding mutant strain of Jinggangmycin was obtained. Enhancing the expression of positive regulatory protein genes can promote the expression of Jinggangmycin biosynthesis genes, and finally significantly increase the production of Jinggangmycin. Compared with the starting strain, the fermentation yield of the high-yielding strain obtained by the present invention is increased by 56%, 18%, 55% and 8% respectively, and the fermentation level of the laboratory shake flask reaches 6.69g/L, 5.05g/L and 6.64g respectively. /L, 4.64g/L.
