abstract The invention discloses a molecular switch for sequence-specific protease detection, and belongs to the field of protein detection. The molecular switch of the present invention includes a HiBiT mutant, LgBiT and SmBiT connected in sequence with a linker; the linker between the HiBiT mutant and LgBiT contains a recognition cleavage sequence. The aforementioned HiBiT mutant has a high affinity for LgBiT, but it has no luciferase function when it is bound to LgBiT, so the molecular switch can have no luciferase activity when it is not in contact with protease, but when exposed to protease, the HiBiT mutant is detached, LgBiT binds SmBiT to produce luciferase activity, which can be detected by a luminescent substrate. The molecular switch of the invention can improve the detection signal-to-noise ratio and has stronger detection sensitivity than the existing molecular switch.
