The invention discloses a recombinant protein vaccine of norovirus GII.4 and enterovirus 71. Taking advantage of the antigen presentation and ability to form viroid particles of the VP1P region of Norovirus while still maintaining good immunogenicity, the main neutralizing epitope site SP70 of EV71 capsid protein VP1 was inserted at its surface loop site 2 , and fused with a histidine (His) tag at the N-terminus to facilitate purification, and the C-terminus is CDCRGDCFC to facilitate the formation of virus-like particles. The codon-optimized gene sequence was cloned into the expression plasmid pET-28a(+) in Escherichia coli High-efficiency expression in BL21 (DE3), a recombinant fusion protein containing both norovirus neutralizing antigen and EV71 neutralizing antigen was prepared as a particle-like vaccine, and the inoculation of mice confirmed its ability to induce specific neutralizing antibody production and immune protection. |
