The present invention relates to the technical field of genetic engineering and bioinformatics, in particular, to a method and application for creating a new gene in an organism without an artificial DNA template. The method is accomplished by simultaneously generating DNA breaks at at least two different specific locations in the genome of an organism, wherein the specific locations are genomic loci capable of dividing different genetic elements or different protein domains, and the DNA breaks are generated by non- Homologous end joining (NHEJ) or homologous repair are used to connect to each other, resulting in new combinations of the different genetic elements or different protein domains that are different from the original genome sequence, forming a new gene. The novel gene of the invention can change the growth, development, resistance, yield and other traits of the organism, and has great application value. |
