abstract | The invention belongs to the field of analytical chemistry, and particularly discloses a method for rapidly determining the content of ustilagin A in urine or a cell culture medium by liquid chromatography-mass spectrometry, which comprises the following steps: (1) sample pretreatment; (2) and (4) detecting by chromatography-mass spectrometry. Aiming at the problem of UA residue analysis in urine and culture medium samples, the method adopts an HLB column to purify and remove coexisting components such as inorganic salt, nonpolar or alkaline micromolecules, and the like, then uses a low-proportion methanol water eluent to enrich and purify through a WAX column, finally successfully eliminates the matrix effect during sample detection by means of an ultra-high performance liquid chromatography-triple quadrupole mass spectrometry combined technology, and realizes the high-sensitivity detection of UA in urine and culture medium for the first time. The method has the advantages of good reproducibility, low sensitivity and the like, the quantitative limit of the ustiloxin A is as low as 50ng/L, and the quantitative detection requirement of the ustiloxin in urine and a cell culture medium can be met. |
