titleOfInvention | Cell-penetrating peptide based on GluN2A subunit and application thereof |
abstract | The invention discloses a cell-penetrating peptide based on GluN2A subunit and application thereof, wherein the cell-penetrating peptide consists of 22 amino acids, and the amino acid sequence is as follows: YGRKKRRQRRRHLFYWKLRFCF are provided. The invention discovers the cell-penetrating peptide 2A-CPP which is derived from a polypeptide sequence designed at the C end of the third transmembrane region of the NMDA 2A receptor for the first time. The polypeptide has better cell membrane penetration capacity under low concentration, interferes the internalization of an NMDA receptor and inhibits ERK phosphorylation due to the specific targeting property of the 2A receptor, can carry biomolecules such as protein and the like into various cells (including adherent culture cells, suspension culture cells, primary culture cells, subculture cells and the like) in vitro through a membrane, and is a transmembrane transport carrier for bioactive molecules such as protein, nucleic acid and the like with great development prospect. Under high concentration, the cell membrane breaking effect can be generated, the half-life period of blood circulation is more than 6h, better cell morphology can be kept, and the sufficient detection of cell surface antigen is ensured. |
