Method and application of a baculovirus expression system for preparing bovine viral diarrhea virus E2 protein |
abstract |
The invention belongs to the technical field of veterinary biological products, in particular to a method and application for preparing bovine viral diarrhea virus E2 protein by a baculovirus expression system. The E2 gene of the BVDV‑LC strain was connected to the pFastBac1 recombinant baculovirus transfer vector, and the recombinant baculovirus vector rBacmid‑E2 was obtained by homologous recombination in E. coli, and the first-generation recombinant baculovirus P1 was obtained after transfection into Sf9 insect cells , continue to infect Sf9 insect cells with the P1 generation virus to obtain the second generation recombinant baculovirus P2 with high infectivity, and then expand the P2 generation virus and purify to obtain the E2 protein. Using the E2 protein prepared by the present invention as a diagnostic antigen and a subunit vaccine can show better advantages, and can be used as a candidate antigen for the development of vaccines and ELISA kits. |
