The invention puts the feed liquid containing amikacin on weak acid cation resin column, then uses 5% -10% ammonium sulfate solution to carry out gradient desorption, according to the different concentration of ammonium sulfate, the impurity with weaker binding force than amikacin is eluted first, then the amikacin sulfate feed liquid is obtained, finally the impurity with stronger binding force is eluted, the obtained amikacin sulfate feed liquid is concentrated and then directly added with methanol to crystallize to obtain amikacin sulfate, the mother liquid is continuously added with methanol to separate out ammonium sulfate, and the ammonium sulfate can be recycled. The ammonium sulfate solution is used as a release agent, the collected amikacin feed liquid is the amikacin sulfate solution, the operation of adding sulfuric acid for salifying is not needed, and the method is simple, green and environment-friendly.
