abstract | The invention discloses an engineering bacterium for efficient fermentation of mixed carbon sources to produce PHB and its application, belonging to the fields of genetic engineering and fermentation engineering. In the present invention, 14 genes of the core sugar gene cluster on the E. coli genome are knocked out to obtain mutant strain WJW01, and 11 genes of the O-antigen gene cluster on the E. coli genome are continuously knocked out to obtain mutant strain WJW02, and then the mutant strain WJW02 is obtained by knocking out 11 genes of the O-antigen gene cluster on the E. The three genes synthesized by PHB were transformed into the streamlined strain WJW02, and the ptsG gene was knocked out, so that the constructed recombinant strain could use the mixed carbon source of xylose and glucose to synthesize PHB up to 78.4% of the dry cell weight, which is the wild-type control strain W3110 /pDXW‑8‑phaCAB (1.5%) and 50% higher than WJW02/pDXW‑8‑phaCAB. |
