abstract | The invention provides a pollen development-related gene, which is the Arabidopsis thaliana orthologous gene AT3G61950 of the turnip pollen development-related gene BrbHLH046 or BrbHLH046. Transcriptome data analysis revealed that BrbHLH046 may be associated with turnip reproductive development. Subcellular localization found that BrbHLH046 is a transcription factor localized in the nucleus. The expression patterns of BrbHLH046 in roots, stems, leaves, flowers, large buds, middle buds and small buds were analyzed, and it was found that the gene had a higher expression level in reproductive organs. The results of promoter activity analysis also showed that BrbHLH046 was highly expressed in the reproductive organs of plants. The ortholog of this gene in Arabidopsis is AT3G61950. The T-DNA insertion homozygous mutant of AT3G61950 was screened by double primer method. The identification of mutant phenotype showed that the pollen abortion rate of Arabidopsis homozygous mutant inflorescence increased after ABA treatment. It is shown that AT3G61950 regulates plant reproductive development in response to hormones, and it also indicates that BrbHLH046 is indeed a candidate gene involved in the reproductive development of turnip, and this gene can be applied to the breeding of horticultural plants such as cabbage. |
