abstract | Firstly, bonding 3,3' -dithiodipropylhydrazide to a chitosan derivative by utilizing a Diels-Alder reaction to obtain a hydrazide chitosan derivative; meanwhile, doxorubicin is connected with 3,3' -dithiodipropylhydrazide through a hydrazone bond to obtain hydrazide doxorubicin; and then uniformly mixing the water solution of the hydrazide chitosan derivative and the hydrazide adriamycin with the platinum drug solution, and forming the core-shell structure type chitosan-based nano prodrug through a self-assembly process. The particle size of the nano prodrug can be adjusted within the range of 100-200nm, the nano prodrug has the dual stimulation responsiveness of glutathione and pH, and the disulfide bond contained in the nano prodrug can eliminate the glutathione, so that the phenomenon that tumor cells tolerate platinum drugs caused by the glutathione is avoided, the nano prodrug can be used for combined chemotherapy of various tumors, the anti-tumor effect is improved, and the toxic and side effects are reduced. |
