abstract | The invention belongs to the field of biotechnology and discloses the application of metallothionein DaMT2a and its encoding gene. The present invention clones the full-length cDNA of the ginseng metallothionein DaMT2a gene for the first time. Gene expression analysis shows that the DaMT2a gene is highly expressed in male flowers and stems, and is up-regulated by ethephon and abscisic acid. At the same time, the gene is induced by low temperature and mechanical injury stress. The expression was up-regulated, and the expression was down-regulated under high temperature stress, so this gene may be closely related to the stress resistance of ginseng and can be used as an important target gene for transgenic breeding of ginseng. The gene was further transferred into the prokaryotic expression strain, and the growth of the transgenic strain under normal conditions, heavy metal ions (Cu 2+ , Cd 2+ ), NaCl stress and hydrogen peroxide stress was significantly improved, so this gene can be used as an important gene. The genetic resources can be applied in the genetic engineering of stress resistance and heavy metal stress resistance of other plants or microorganisms other than ginseng potato. |
