abstract The invention discloses a method for rooting and rooting smilax glabra tissue culture seedlings, which comprises the following steps: firstly, cutting the glabrous greenbrier rhizome proliferation cluster buds into 3-5 buds serving as small clusters on a superclean workbench, and inoculating the small clusters into a strong bud culture medium for culture to obtain thick and strong cluster buds; and cutting the obtained robust cluster buds into single buds, inoculating the single buds into a rooting rhizome induction culture medium for culture, wherein when 55-65 days are needed, the number of the roots of each seedling is 2-10, the number of the rhizome is 1-7, the average number of the roots of each seedling in different treatments is 2.79-7.33, and the average number of the rhizome of each seedling is 1.53-5.46. The method can directly root the smilax glabra bud and simultaneously knot the rhizome, one formula induces two organs of the root and the rhizome, and the rhizome is a medicinal organ of the smilax glabra, so that the transplanting survival rate of the smilax glabra is improved, and the aim of improving the yield of the smilax glabra is fulfilled by increasing the number of the rhizome. The method has the advantages of simple operation, good repeatability, cost reduction, safety, high efficiency and the like.
