abstract |
The present disclosure relates to systems, devices, and methods for nucleic acid sequencing, including polynucleotide strands having nucleotides modified with redox labels attached thereto or capable of receiving modified nucleotides having redox labels attached thereto. The systems, devices, and methods include a dielectric member having an attached translocator protein located between oxidation and reduction electrodes. The oxidation and reduction electrodes generate an electric field that extends to a reaction region where translocation of the polynucleotide chain through the protein occurs, thereby identifying the modified nucleotide having the redox label attached thereto by a change in current in the oxidation and reduction electrodes, wherein the change identifies electron transfer from the reduction electrode to the redox label and to the oxidation electrode when the redox-labeled modified nucleotide of the polynucleotide chain having a nucleobase covalently bonded to the modified nucleotide is in the reaction region. |
