abstract The invention discloses a construction method of a standardized cord blood hematopoietic stem cell bank. The construction method of the standardized cord blood hematopoietic stem cell bank comprises the following steps: extracting umbilical Cord Blood Hematopoietic Stem Cells (CBHSC) from umbilical cord blood by using a self-made stem cell separation solution, wherein the umbilical cord blood is subjected to ABO/Rh blood type, HLA typing and microbiological detection before extraction; and performing relevant quality detection on the obtained CBHSC, treating the CBHSC meeting the quality standard by using a cryoprotectant, subpackaging the CBHSC into a freezing tube or a freezing bag, performing gradient cooling, performing liquid nitrogen freezing preservation according to the ABO/Rh blood type and HLA type of the CBHSC, and establishing a CBHSC information file which can be retrieved, namely a standardized CBHSC library. Compared with the conventional umbilical cord blood bank, the standardized CBHSC bank constructed by the method has high cell resuscitation survival rate, better hematopoietic reconstruction function and less erythrocyte residue, can improve the success rate of stem cell transplantation and guide the formulation of a stem cell treatment scheme of related diseases or the research of stem cell medicines.
