abstract The present invention provides a preparation method of core-shell nucleic acid-immobilized microspheres, which can effectively immobilize nucleic acid molecules. The nucleic acid-immobilized microspheres of the present invention have good singleness, and are favorable for applications in gene sequencing and the like. By connecting initiators or carbon-carbon double bonds on the surface of the microspheres; then in the aqueous phase, the microspheres are polymerized to connect polymer chains containing click chemistry functional groups and azide groups on the surface; and then the nucleic acid is immobilized by click chemistry molecular. The steps are simple and can be applied to common sequencers.
