The amino acid sequence of the nicotinamide ribokinase mutant is compared with the wild-type nicotinamide ribokinase whose amino acid sequence is shown in SEQ ID NO: 2. The 102nd, 52nd, and 146th positions were mutated by any one of single mutation, pairwise combined mutation, three combined mutation or four combined mutation. The nicotinamide ribokinase mutant provided by the present invention can be used for the synthesis and preparation of β-nicotinamide mononucleotide. Compared with the wild-type enzyme, the nicotinamide ribokinase mutant constructed by the invention has the enzyme activity increased by 3.86-4.53 times, shortens the reaction time, can significantly reduce the usage amount of the enzyme, reduces the fermentation capacity and cost, improves the NMN yield, and has the advantages of Broad prospects for large-scale industrial applications. |
