abstract |
A "binary differential" method for finding a combination of small exquisite DNA methylation positions (CG IDs) in the human genome to detect cancer in DNA in patient-derived biological material and distinguish it from other non-tissue cell DNA and blood cell DNA. Another method for detecting the tissue of origin of tumor DNA uses a combination of unique DNA methylation positions (CG IDs) in the human genome. Combinations of CG IDs derived from tumor DNA are disclosed for accurately detecting cancer by measuring DNA methylation for combinations of specific CG IDs and deriving a "methylation score". A kit for predicting cancer using CG ID using multiplex next generation sequencing methylation assays, pyrosequencing assays and methylation specific PCR in small plasma. Various methods of using biological materials help predict cancer in humans who do not have other clinical signs of cancer. |
