abstract The invention discloses a DNA frame wrapped G-quadruplex structure and a preparation method and application thereof. The invention mixes 6 DNA sequences needed by assembling a framework structure, a Cy3 sequence and a Cy5 sequence needed by forming a G-quadruplex probe in a buffer solution with a final concentration of 0.6 mu M uniformly, puts a sample into a PCR instrument, and anneals the sample from 95 ℃ to 4 ℃ within 3.5 hours to obtain the G-quadruplex nanostructure (G4-F) wrapped by the DNA framework, and only generates green fluorescence from Cy3 under the condition that a target sequence (miR-122a) is deleted. When the target sequence exists, the fluorescence resonance energy transfer process can be promoted, so that the fluorescence of Cy3 is weakened and the fluorescence of Cy5 is strengthened. The preparation method is simple and low in cost, the prepared G4-F can specifically detect the target, is not interfered by external environmental factors, has high stability, and the DNA framework structure can effectively protect the G-quadruplex probe from being degraded by intracellular substances in cells, and can efficiently identify the target sequence and realize intracellular imaging detection.
