titleOfInvention | A kind of recombinant Escherichia coli for efficiently producing glutaric acid and construction method thereof |
abstract | In the present invention, the lysine decarboxylase gene cadA, the butanediamine aminotransferase gene patA and the γ-aminobutyraldehyde dehydrogenase gene patD are cloned from the genome of Escherichia coli MG1655 by means of molecular biology, and obtained from Pseudomonas fluorescens Pseudomonas The 4-aminobutyric acid transaminase gene gabT and the succinic semialdehyde dehydrogenase gene gabD were cloned from the fluorescens genome. After the constructed recombinant expression vector pETM6R1‑cadA‑RBS01‑patA‑RBS02‑patD‑gabT‑gabD was introduced into E. coli W3110, the recombinant strain Glu‑02 that efficiently produced glutaric acid was obtained by ampicillin resistance plate screening . The recombinant strain adopts a fed-batch fermentation strategy in a 5-L fermentor, and is fermented for 60 hours, the yield of glutaric acid reaches 56.2 g/L, and the conversion rate is 62.2%. |
