abstract The invention relates to a calixarene-GCA-cyclodextrin-CD co-assembly and its application in the preparation of medicines. The co-assembly is constructed by GCA and CD at a molar ratio of 1:1 through weak π-π interactions and hydrophobic interactions, which can achieve selective and strong bonding with β-amyloid through heteropolyvalent interactions, and can inhibit β-amyloid fibrilization, depolymerization of β-amyloid fibrils, reduction of β-amyloid oligomer content, and alleviation of the cytotoxicity of β-amyloid fibrils. The guanidine group on the surface of the co-assemblies facilitates the phagocytosis of β‑amyloid by microglia. Animal experiments have shown that the co-assembly can reduce the content of soluble and insoluble β-amyloid and β-amyloid oligomers in the brains of Alzheimer's mice, and can improve the Cognitive impairment.
