The present invention provides a fourth-generation CAR-T cell and an application thereof, wherein the fourth-generation CAR-T cell expresses a chimeric antigen receptor that specifically binds to an antigen, secreted IL-7 and NFAT-regulated CCL19; The signal peptide of the IL-7 is the IL-10 signal peptide; the promoter of the CCL19 is the NFAT regulated promoter. In the present invention, the original signal peptide of IL-7 is changed to the signal peptide of T cell secretory protein IL-10, and the NFAT regulated promoter is used as the promoter of CCL19, so that CAR-T cells can secrete IL-7 into the CCL19. The extracellular and conditional expression of CCL19 not only promotes the proliferation of CAR-T cells, but also recruits immune cells to the tumor to the greatest extent, significantly improving the anti-tumor efficacy of CAR-T. |
