abstract The invention discloses a manganese ion-independent mutant of manganese peroxidase and its application. The present invention carries out site-directed mutation to the 70th amino acid residue Arg and the 166th amino acid residue Glu on the basis of the I1-MnP1 amino acid sequence, and through constructing the expression strain, inducing expression, denaturing and renaturing and purifying, the mutant MnP is obtained (R70V/E166A). The mutant of the present invention can effectively decolorize anthraquinone dyes in a neutral pH environment independent of Mn 2+ , and has an improved tolerance to H 2 O 2 concentration, which is not effective for water bodies caused by anthraquinone dyes. Pollution control is of great significance.
