The present disclosure provides methods for preparing a sequencing library comprising the steps of providing a template nucleic acid sequence, dNTPs, dUTPs, primers, polymerases, dUTP excisionases, and a plurality of oligonucleotide adapter sequence segments comprising beads; amplifying the template nucleic acid with the polymerase, dNTPs, dUTPs, and random hexamers to provide complementary nucleic acid sequences comprising occasional dUTPs; and excising the incorporated dUTPs with the dUTP excision enzyme to A nick is provided in the complementary nucleic acid sequence, thereby providing a sequencing library. |
